Hello Callum, The product you have posted the question on is just the speaker itself and not an amplifier kit, we do sell a kit that uses this speaker https://www.kitronik.co.uk/2158-3w-class-d-stereo-amplifier.html, this contains all the parts for the amplifier including two speakers.
Will this work with the MP3 Mono Amplifier V2.0 Kit?

These are the speakers that are supplied with the 3W class D amp kit, so yes they are fine to use with it.

The amplifier chip used in the high powered amp kit is not specified to drive a 4 Ohm load, as such you may have issues with using these speakers. We would suggest using an 8 Ohm speaker as this is what the amplifier chip is designed to drive.
Please can you confirm if these speakers are suitable to operate in a car on dc 12 volt?

It depends on the amplifier in your car.What matters is that the amplifier can support the impedence of the speakers (4 Ohm). If it can't it may damage the amplifier or speaker.
